The collision occurred around 10.30am on the main N25 road near Gaulstown, Glenmore, Co Kilkenny.
The woman was pronounced dead at the scene, and has been removed to a mortuary for a post-mortem examination.
Gardaí and emergency services attended the scene on the stretch of N25 road, which is now closed as Garda Forensic Collision Investigators carry out a technical examination.
Meanwhile in a separate collision, a teenage girl died after the car she was driving left the road at Kilaldry Bridge in south Tipperary and entered the River Aherlow last night.
Local politicians in Tipperary have said the community is distraught at the loss of the local girl, who is understood to have been due to sit her leaving cert this summer.
Gardaí and emergency services attended the scene of the single-vehicle crash, which occurred at Kilmoyler in Cahir after 10pm on Thursday.

Cathaoirleach of Tipperary County Council, Declan Burgess, said the N24 road is regarded as dangerous road, and the road the crash happened on is a small local road off the N24.
“People are very upset out there. An 18-year-old girl, it’s incredibly said,” said Cllr Burgess. “It’s horrible news to wake up to this morning alright. Awful, awful.”
The body of the girl, who was the driver and sole occupant of the car, was recovered from the water this morning by members of the Garda Water Unit and has been removed from the scene to the mortuary where a post mortem examination is due to take place.
The L3101 is currently closed and local diversions are in place. A technical examination is being conducted by Garda Forensic Collision Investigators at present.
Local councillor John O’Heaney said it was “a very sad and tragic day here in Tipperary.”
“I would like to extend my deepest sympathies to the family and friends of this lady,” he told RTÉ Radio One. “It’s a very sad day for everyone.”
Councillor John Fitzgerald said there was “absolute heartbreak” in the community.
“A cloud has descended on us today. It’s just awful really, really terrible news,” he told RTÉ.
Mr Fitzgerald said he knew the girl’s family and that she was “a beautiful girl, a really good person”.
“The community and all in Clonmel here are in shock,” he said.
Gardaí are appealing to anyone who may have witnessed the incident or were in the Kilmoyler area to please contact them.
“Any road users who may have camera footage (including dash-cam) and were travelling on the L3101 or in the Kilmoyler area between 10pm and midnight last night are asked to make this available to investigating gardaí,” they added.
“Anyone with information asked to contact Cahir Garda Station on 052 7445630, the Garda Confidential Line on 1800 666 111, or any Garda station.”
